Best Biotech Companies To Invest In Right Now

Health care analyst Max Macaluso recently sat down with Fool contributor Keith Speights to talk about some of the best speculative biotech plays on the market. Max and Keith discussed three companies that investors should consider: Keryx Pharmaceuticals (NASDAQ: KERX  ) , Sarepta Therapeutics (NASDAQ: SRPT  ) , and MannKind (NASDAQ: MNKD  ) .

Both Keryx and Sarepta have soared over the last year. Keryx is up a whopping 190% so far in 2013. Meanwhile, Sarepta's shares jumped nearly 50% year-to-date after an amazing run last year.

The nice run by Keryx was powered by high expectations for its kidney disease drug Zerenex. Sarepta's success has stemmed from excitement over its Duchenne muscular dystrophy drug eteplirsen.

MannKind also has enjoyed a great year so far. Shares are up nearly 80% in the first three and a half months of 2013. Investors are eagerly awaiting results from what they hope to be the final study for inhalable insulin product Afrezza.

Protalix BioTherapeutics, Inc. is a biopharmaceutical company focused on the development and commercialization of recombinant therapeutic proteins based on its ProCellEx protein expression system, ProCellEx. Its oral taliglucerase alfa is a plant cell expressed form of GCD that is naturally encapsulated within carrot cells genetically engineered to express the GCD enzyme. Pre-clinical studies of oral taliglucerase alfa demonstrate the stability of the enzyme in the cell and the capacity of the cell�� cellulose wall to protect the enzyme against degradation in the digestive tract in an in-vitro model of the stomach and intestines. Additionally, rats fed with lyophilized carrot cells expressing GCD have accumulated the active enzyme in the target organs; the spleen and liver. As of December 31, 2010, the Company had completed Phase III Clinical Trial.

PRX-102

The Company is developing PRX-102, its plant cell expressed modified version of the recombinant ! alpha-GAL-A protein, a therapeutic enzyme for the treatment of Fabry disease. Fabry disease is a rare, hereditary, genetic lysosomal storage disorder in humans caused by an X-lined deficiency of the alpha-GAL-A enzyme. The Company is in the animal evaluation testing phase of the development of PRX-102, which tests are based on a mouse model for Fabry disease.

Acetylcholinesterase

Protalix Ltd. is a wholly owned subsidiary of the Company is licensed the rights to certain technology under a research and license agreement with Yissum Research and Development Company (Yissum) and the Boyce Thompson Institute, Inc. Pursuant to the agreement, the Company is developing PRX-105, a plant cell-based acetylcholinesterase (AChE) and its molecular variants for the use in several therapeutic and prophylactic indications, as well as in a biodefense program and an organophosphate-based pesticide treatment program.

As of December 31, 2010, its in-vitro experiments of PRX-105 have shown that the acetylcholinesterase enzyme in its ProCellEx protein expression system demonstrates biological activity on biochemical and cellular levels. In addition, early animal studies demonstrated that the acetylcholinesterase expressed in its ProCellEx protein expression system was able to treat animals exposed to the nerve gas agent analogues, both when injected with its acetylcholinesterase product candidate immediately before exposure or when injected after exposure. In March 2010, it initiated a preliminary phase I clinical trial of PRX-105, which the Company completed in June 2010.

pr-antiTNF

pr-antiTNF is a candidate for the treatment of certain autoimmune diseases such as rheumatoid arthritis, juvenile idiopathic arthritis, ankylosing, spondylitis, psoriatic arthritis and plaque psoriasis. The Company has designed the antiTNF as pr-antiTNF. pr-antiTNF is a plant cell-expressed recombinant fusion protein made from the soluble form of the human TNF receptor (TNFR), f! used to t! he Fc component of a human antibody domain. pr-antiTNF has an identical amino acid sequence to Enbrel and its in-vitro and preclinical animal studies have demonstrated that pr-antiTNF exhibits similar activity to Enbrel. Specifically, pr-antiTNF binds TNF thereby inhibiting it from binding to cellular surface TNF receptors and protects L929 cells from TNF-induced apoptosis in a dose-dependent manner.

    Shares of Gilead (GILD) and Celgene (CELG) have tumbled today after both companies revealed that they were facing patent pressures.

    For Gilead, Idenix Pharmaceuticals (IDIX) filed a patent-infringement lawsuit that appears to target Gilead’s Sovaldi (someone please correct me if I’m wrong). For Celgene, it was the decision of a New Jersey court to schedule a hearing on a patent case for April 29, earlier than expected. It hasn’t helped that the UK’s National Institute for Health and Care Excellence also said that Celgene’s Revlimid doesn’t work well enough to be covered for “second line treatment of multiple myeloma.”

    Bernstein’s Geoffrey Porges doesn’t expect the outcome of Celgene’s hearing to have a major impact:

    Overall, the outcome of the case will likely not be impacted by the outcome of the Markman hearing. Revlimid is currently protected by some two dozen patents in the US. Our attorney consultants believe that the polymorph patent 7465800 (or ’800 patent) is a particularly solid pillar in the “dome” of patents that Celgene has built around Revlimid. Our outlook for Revlimid sales remains unchanged, we continue to project product sales growth from $9bn in 2015 to $16bn in 2019, and $21bn 2023. Based on our analysis of consensus expectations 2, we estimated that the stock had been already discounting a ~30% chance that Revlimid will lose its patent protection by 2019…

    We continue to rate Celgene Outperform, with a target price of $214. We believe that the market’s concern about Revlimid’s patent protection and NICE recommendations is overdone. We see no reason to change our revenue and earnings forecasts for the stock and recommend investors add to positions on this temporary weakness.

Five Prime Therapeutics, Inc., incorporated on December 20, 2001, is a clinical-stage biotechnology company focused on discovering and developing protein therapeutics. Protein therapeutics is antibodies or drugs developed from extracellular proteins or protein fragments that block disease processes, including cancer and inflammatory diseases. The Company�� advanced product candidates include FP-1039/GSK3052230 (FP-1039), FPA008 and FPA144. FP-1039 is a protein therapeutic that traps and neutralizes cancer-promoting fibroblast growth factors (FGFs), involved in cancer cell proliferation and new blood vessel formation. FP-1039

FP-1039 is a protein therapeutic, which includes the extracellular part of FGFR1. FP-1039 acts as an inhibitor of FGFs, because the FGFR1 portion of the molecule binds to FGFs and prevents them from binding to FGFR1 on tumor and blood vessel cells. Because FGF proteins circulating in the blood are called ligands, FP-1039 is called a ligand trap. FP-1039 also includes a portion of an antibody called the Fc region. In preclinical testing, it observed inhibition of tumor growth with single-agent FP-1039, particularly in tumors withFGFR1 gene amplification, including squamous NSCLC and SCLC.

FPA008

FPA008 is an antibody that inhibits CSF1R and is being developed to treat patients with RA. FPA008 also has the potential to treat patients with other inflammatory diseases, including lupus nephritis, psoriatic arthritis, ankylosing spondylitis, fibrosis, inflammatory bowel disease and multiple sclerosis. These are chronic, incurable disorders with serious medical complications and disability for ! which better therapies with novel mechanisms of action are needed. FPA008 is an anti-CSF1R antibody, which it designed to block the ability of IL-34 and CSF1 to bind to and activate CSF1R. FPA008 reduces the numbers and activity of monocytes and macrophages that cause disease, and prevents the production and release of inflammatory factors. The Company and others has demonstrated that both IL-34 and CSF1 are present at increased levels in the inflamed joints of patients with RA.

FPA144

FPA144 is a monoclonal antibody directed against a form of FGFR2, or FGFR2b. When the FGFR2 gene is amplified by cancer cells, the FGFR2b protein is expressed at abnormally high levels on the tumor�� surface. This occurs in some patients with gastric and lower esophageal cancers. The tumor cells that have too much FGFR2b protein on their surface can be identified by special staining tests performed on the tumor. Because FGFR2b is the target for FPA144, patients��tumors can be screened for this protein, helping to identify the patients most likely to respond to FPA144 treatment.

Mast Therapeutics, Inc., formerly ADVENTRX Pharmaceuticals, Inc., incorporated in December 1995, is a development-stage company biopharmaceutical company focused on developing product candidates. The Company's product candidate is ANX-188, a rheologic, antithrombotic and cytoprotective agent that improves microvascular blood flow and has application in treating a range of diseases and conditions, such as complications arising from sickle cell disease. As of December 31, 2011, the Company also is developing ANX-514, a detergent-free formulation of the chemotherapy drug docetaxel. The Company�� purified form of P188 (purified P188), which is the active ingredient in ANX-188, was designed to eliminate certain low molecular weight substances present in P188 (non-purified), which is primarily responsible for the moderate to moderately severe elevations in serum creatinine levels (acute renal dysfunction) observed in prior clinical studies of P188 (non-purified). Purified P188 has been evaluated in multiple clinical studies by a prior sponsor, including a 255-patient, phase III study.

ANX-514 (docetaxel for injectable emulsion)

ANX-514 is a detergent-free emulsion formulation of docetaxel, an intravenously-injected chemotherapy drug commonly used to treat solid tumors. Taxotere, a branded form! ulation of docetaxel, is approved to treat breast, non-small cell lung, prostate, gastric, and head and neck cancers. ANX-514 was designed to have clinically comparable release of docetaxel relative to Taxotere while eliminating the presence of polysorbate 80 and ethanol, both of which are used to solubilize docetaxel in the Taxotere formulation. The ANX-514 formulation solubilizes docetaxel using oil droplets consists of a combination of non-toxic excipients. Docetaxel is contained within these oil droplets and can be administered intravenously without using detergents as pharmaceutical vehicles. Once in central circulation, the emulsion is metabolized rapidly, leaving chemically-identical active ingredient to exert its cytotoxic effect. ANX-514 may reduce the incidence and severity of hypersensitivity reactions and delay the onset of fluid retention.

Exelbine (vinorelbine injectable emulsion)

Exelbine is an emulsion formulation of the chemotherapy drug vinorelbine. Navelbine, a branded formulation of vinorelbine, is approved in the United States to treat advanced non-small cell lung cancer as a single agent or in combination with cisplatin, and approved in the European Union to treat non-small cell lung cancer and advanced or metastatic breast cancer.

In August 2011, the Company received a complete response letter from the Food and Drug Administration (FDA) stating that it could not approve the Exelbine Non Disclosure Agreement (NDA) in its present form and that the bioequivalence study would need to be repeated because the authenticity of the drug products used in the bioequivalence trial could not be verified in accordance with FDA standards. However, the Company elected to discontinue independent development of Exelbine and as of December 31, 2011, the Company was seeking a partner or outside investor for the program to complete the necessary bioequivalence study.

    Mast Therapeutics (MSTX), a biopharmaceutical company, focuses on developing therapies for serious or life-threatening diseases. This stock closed up 2.1% 65 cents per share in Thursday's trading session.

     

    Thursday's Range: $0.63-$0.67

    52-Week Range: $0.40-$1.10

    Thursday's Volume: 1.11 million

    Three-Month Average Volume: 1.88 million

     

    From a technical perspective, MSTX spiked modestly higher here right above its 50-day moving average of 61 cents per share with decent upside volume. This stock has been trending sideways and consolidating for the last month, with shares moving between 60 cents on the downside and 72 cents on the upside. Shares of MSTX are now starting to push higher and into range of triggering a big breakout trade above the upper-end of its recent sideways trading chart pattern. That trade will hit if MSTX manages to clear some near-term overhead resistance levels at 68 to 72 cents per share with high volume.

     

    Traders should now look for long-biased trades in MSTX as long as it's trending above some key near-term support at 60 cents per share and then once it sustains a move or close above those breakout levels with volume that hits near or above 1.88 million shares. If that breakout materializes soon, then MSTX will set up to re-test or possibly take out its next major overhead resistance levels at 80 to 85 cents per share, or even 93 cents per share.

    A little over a month ago, owning Mast Therapeutics, Inc. (NYSE: MSTX) was nothing but pure misery. Shares plunged from $0.63 to $0.43 in one day, when details of a dilutive public offering were unveiled. As is so often the case though, the market may have overshot with the selling effort that KO's MSTX. Though putting more shares 'out there' mathematically meant existing shareholders would have to share more of the company's upside with newcomers, what's slowly coming to light is that the inflow of new cash is still more advantageous to those prior shareholders; the company would progress little without it.
